- PublicaciónAcceso abiertoMedición de β-hCG procedente de líneas celulares de coriocarcionoma humano JEG-3 BeWo por QLIA(2019-10-28) Ocampo Duque, Luz Fanny; Sánchez Arredondo, Sandra Patricia; Quintana Castillo, Juan CarlosIntroduction: The cell lines JEG-3 and BeWo are clonal derivates of choriocarcinoma of human placenta and share some properties of immature human trophoblast cells, including the secretion of β-hCG, and thus they have been proposed as a trophoblast producer of hormones model. Beta subunit (β-hCG) is usually measured in clinical laboratory using the QLIA immunoassay method. Objective: The objective of this study is to evaluate the use of the automated IE technique in the quantification of the β-hCG produced from a culture system verifying potential interferents when a measurement matrix, supported by the clinical laboratory, is changed. Materials and Methods: Cell lines JEG-3 and BeWo were cultured in RMMI 1640 with the aim of establishing the capacity to produce β-hCG after 48 hours of culture compared witgh serums of normal pregnant women. To determine the concentration of the hormone, we used the Integrated System VITROS 5600 and the reactive Total β-hCG II of the clinical laboratory Ortho Clinical Diagnostics utilizing serums of pregnant women with different concentrations of β-hCG. Results and Conclusions: A low rate of recovery of β-hCG measured from cell cultures was obtained probably due to phenol red which could be one of the most involved interferents in the measurement.
